Jon Robertus once told me that there are no longer any more crystallographers, just people who press buttons. In one sense he’s absolutely right. Investigators no longer build atomic models by hand using mirrors and photographic film. I would only add to this point that successful structural biologists know which buttons to press and why. My goal with this section is to collate knowledge regarding some of the core fundamentals regarding cryo-EM. We start first with the physics, hardware, and mathematics behind cryo-EM. From here I’ll briefly discuss sample application for SPA, which then naturally leads to data collection and image processing. All that is left from this is a discussion of reconstructions and validations.
Obviously, a significant undertaking that will continuously be updated and modified.
