SOB (Super Optimal Broth) Media

From Cold Spring Harbor Protocols

Dissolve the following in 800mL of distilled water

  1. 20g Tryptone
  2. 5g yeast extract
  3. 0.584g sodium chloride (10mM)
  4. 0.186g Potassium Chloride (2.5mM)
  5. 2.4g MgSO4 (20mM)

Volume up to 1L. Autoclave. Some protocols advise to adjust pH to 7, however if Doug Hanahan did not bother then why should we?

Make SOB as before. Let cool to 60C or less, and then add 20mL of a 1M glucose solution.

Glucose solutions can be autoclaved for very short periods of time, however it is sufficient to make the glucose solution and filter using a 0.22um filter.

LB Media

Makes 1L.

  1. Add 10g of tryptone to 800mL of water while stirring.
  2. Add 10g of NaCl
  3. Add 5g of yeast extract

Stir until dissolved. Volume up to 1L and then autoclave.
