Preparation of Dialysis Tubing

Taken from p456 of "Molecular Cloning: A Laboratory Manual" by Maniatis et al.

  1. Cut the dialysis tubing into pieces of convenient length (10-20cm). Use half the roll of dialysis tubing.
  2. Boil for 10 minutes in a large volume of 2% sodium bicarbonate and 1mM EDTA. Boil using the microwave, having the tubing and solution submerged in a large beaker, with a smaller beaker on top to hold the dialysis tubing down (as the dialysis tubing will float).
    1. The 600mL beaker and the 250mL beaker work well for this task
    2. The volumes needed are just enough to cover the tubing (~300 generally, so 6g Sodium Bicarb+600uL EDTA)
  3. Rinse the tubing thoroughly in distilled water.
  4. Boil for 10 minutes in 0.001M EDTA (600uL of 0.5M for 300mL).
  5. Allow to cool and store in 20% EtOH @5C. Be sure that the tubing is always submerged.
  6. Before use, wash the tubing inside and out with distilled water, always handle the tubing with latex gloves.
